实验原理

① 35 μL外泌体样品滴加在芯片上孵育;

② 预包被的抗体异结合外泌体表面蛋白以捕获外泌体;

③ 再使用荧光抗体异性标记需要表征的标记物;

④ 后用ExoView R100检测外泌体粒径、计数、蛋白表达(CD9,CD81,CD63等)及共定位。

芯片上可包被多达6种捕获抗体,异性捕获含定蛋白标记物的外泌体。

对阳性外泌体计数

芯片捕获外泌体后,可通过SP-IRIS技术直接检测样品中外泌体的数量。

 

 

单个外泌体蛋白共定位分析

检测每个外泌体的荧光信号并进行统计,可获得荧光共定位信息,用于分析样品中不同表型外泌体的比例(如右图所示)。

 

 

无需纯化

 

使用抗体捕获模式,防止样品中杂质影响结果,可直接检测血液、尿液和细胞培养液中的外泌体,未纯化样品的测量结果与纯化后基本致(如右图所示)。

 

 

粒径分辨率高

 

高精度SP-IRIS技术,可检测≥50 nm的外泌体,测量结果与电子显微镜检测结果基本致,并统计生成外泌体的粒径分布结果(如右图所示)。

 

 

可检测外泌体内容物

 

试剂盒配套相应的穿膜剂,可穿透外泌体并对外泌体内容物进行染色并检测,未穿膜时只能检测到跨膜蛋白CD9的荧光信号,穿膜后即可检测到外泌体内容物Syntenin的表达(如右图所示)。
